چکیده
Native mass spectrometry is now an important tool in structural biology. Thus, the nature of higher protein structure vacuum spectrometer area significant interest. One major goals study gas-phase to elucidate stabilising role interactions at level individual amino acid residues. A strategy combining chemical modification together with collision induced unfolding (CIU) was developed and employed probe compact ions produced by native electrospray ionisation. Tractable used alter properties residues, ion mobility-mass (IM-MS) utilised monitor extent as a function modification. From these data importance specific intramolecular for stability can be inferred. Using this approach, aided molecular dynamics simulations, interaction between K6 H68 ubiquitin identified, contact N-terminus E22 binding UBA2.

Snowflake Vitreoretinal Degeneration (SVD) Mutation R162W Provides New Insights into Kir7.1 Ion Channel Structure and Function
Snowflake Vitreoretinal Degeneration (SVD) is associated with the R162W mutation of the Kir7.1 inwardly-rectifying potassium channel. Kir7.1 is found at the apical membrane of Retinal Pigment Epithelial (RPE) cells, adjacent to the photoreceptor neurons. The SVD phenotype ranges from RPE degeneration to an abnormal b-wave to a liquid vitreous. We sought to determine how this mutation alters the...
متن کاملLigand binding and unfolding of tryptophan synthase revealed by ion mobility-tandem mass spectrometry employing collision and surface induced dissociation
Understanding protein tertiary and quaternary structures is crucial to a better understanding of their biological functions. Here we illustrate for tryptophan synthase that tandem mass spectrometry (MS/MS) reveals not only protein subunit architectures, but also protein unfolding behavior when coupled with ion mobility (IM). In the present study, we verified the subunit arrangement with surface...
متن کاملComparing equilibrium and kinetic protein unfolding using time-resolved electrospray-coupled ion mobility mass spectrometry.
Protein unfolding intermediates are thought to play a critical role in conformational pathogenesis, acting as a 'gateway' to inactivation or pathogenic aggregation. Unfolding intermediates have long been studied either by populating partially-folded species at equilibrium using incresingly denaturing conditions, or by transiently populating 'kinetic' intermediates under fully denaturing conditi...
متن کاملProtein Unfolding and the Diffusion Collision Model
In the diffusion-collision model, the unfolding or backward rates are given by the likelihood of secondary structural cluster dissociation. In this work, we introduce a backward rate calculation modeled from a Kramers-type thermal tunneling through a barrier, which represents the free energy potential well for buried hydrophobic residues. Our results are in good agreement with currently accepte...
متن کاملUnfolding the Unfolded Protein Response: Unique Insights into Brain Ischemia
The endoplasmic reticulum (ER) is responsible for processing of proteins that are destined to be secreted, enclosed in a vesicle, or incorporated in the plasma membrane. Nascent peptides that enter the ER undergo a series of highly regulated processing steps to reach maturation as they transit the ER. Alterations in the intracellular environment that induce ER stress are thought to interrupt th...
